Repurposed from its original role as an SSRI for premature ejaculation, Dapoxetine has emerged as an inhibitor of ZIKV replication. It binds selectively to the RdRp “N” pocket, a critical RNA synthesis site. SPR analysis showed a Kd of 49.8 μM. In vitro studies across multiple cell lines confirmed inhibition of viral RNA, proteins, and infectious titers, with EC50s in the low micromolar range. Its low cytotoxicity and favorable clinical profile suggest it could be used against ZIKV and other RNA viruses.
