Melanotan-II (MT-II) is a synthetic cyclic peptide related to α-MSH, originally developed for skin tanning. It activates melanocortin receptors, especially MC4R, leading to central initiation of erections. Unlike traditional ED drugs, it works via the brain and spinal cord rather than penile tissue. MT-II was effective in animal and human trials, showing erection rates up to 80%. Nitric oxide appears to mediate its peripheral results. Its structure allows CNS penetration and resistance to degradation, making it a strong candidate for sexual dysfunction treatment.
