Initially approved as a selective serotonin reuptake inhibitor (SSRI) for premature ejaculation, Dapoxetine has been repurposed as a candidate inhibitor of Zika virus RNA polymerase. It binds specifically to the RdRp “N” pocket, essential for RNA initiation. Surface plasmon resonance confirmed this interaction with a Kd of 49.8 μM. In vitro studies across various cell lines showed potent ZIKV suppression, with low EC50 values and minimal toxicity. These findings, along with its favorable safety and pharmacokinetics, support further antiviral investigation.
