Melanotan-II (MT-II), originally developed as a tanning agent in the 1980s, demonstrated unexpected pro-erectile effects when a test subject unintentionally overdosed. MT-II is a potent activator of MC receptors, particularly MC4R, making it "superpotent" compared to natural peptides like α-MSH. MT-II works centrally through both forebrain and spinal mechanisms, stimulating erections via pathways that ultimately involve nitric oxide for vasodilation in peripheral tissues.
