Melanotan-II (MT-II) was synthesized to enhance skin pigmentation but unexpectedly revealed strong proerectile effects during clinical trials. Its structure, based on α-MSH and ACTH, allows it to cross the blood-brain barrier and activate MC4 receptors associated with sexual arousal and penile erection. In studies involving men with psychogenic and organic erectile dysfunction, MT-II effectively induced erections, with increased rigidity and duration compared to placebo. Despite its efficacy, side effects like nausea and appetite suppression were dose-dependent, highlighting the need for cautious dosing.
