The development of flurbiprofen–antioxidant mutual prodrugs combines effective anti-inflammatory properties with improved gastrointestinal safety. Using natural antioxidants as esterifying agents, the prodrugs temporarily mask the carboxylic acid group, mitigating direct gastric irritation. Upon hydrolysis in plasma, they release active flurbiprofen. These prodrugs demonstrated strong anti-inflammatory and analgesic effects, with reduced gastric toxicity in animal studies. Computational analyses confirmed selective COX-2 inhibition, supporting their potential as safer NSAIDs.
