Tadalafil citrate, known as Cialis, was developed by ICOS Corporation in Washington and later co-marketed by Eli Lilly. Initially studied as IC351 for cardiovascular use, it was repurposed as a PDE5 inhibitor for erectile dysfunction (ED). Approved by the FDA in 2003, it offered a major advantage with its 36-hour duration, attributed to its unique piperazinedione structure. Unlike sildenafil, tadalafil has a longer half-life of 17.5 hours, allowing for extended efficacy and more spontaneous sexual activity .
